Cryptic genetic variation shapes the fate of gene duplicates in a protein interaction network
Introduction
Proteins evolve in a vast sequence space1 that contains many functionally similar proteins. Proteins can, therefore, change in sequence while maintaining their function2. Such changes in sequence that do not alter function constitute cryptic genetic variation3. Although molecular functions are preserved when a given protein explores this space, its ability to evolve in the future could change. The accumulation of cryptic mutations could create a genetic context where the same mutation will have different effects on functionally equivalent proteins due to epistasis4,5. Cryptic variation and epistasis, therefore, dictate whether a mutation in functionally similar proteins will be adaptive, neutral or deleterious6,7,8,9.
One demonstration that such cryptic variation impacts evolution is that many substitutions introduced in orthologous proteins performing similar functions can have opposite effects (neutral or adaptive in one and deleterious in the other)10,11. Epistasis determines the effect a mutation has on protein function by making it contingent on the genetic background of such proteins5,12. Just as cryptic variation impacts how orthologs respond to
